    350 (BTBR x B6.Cg-Lepob)F2 animals were typed at 99 polymorphic loci to identify QTLs modifying obesity. BTBR.Cg-Lepob/Lepob animals are severely obese and exhibit significantly increased food intake compared to B6.Cg-Lepob/Lepob animals.

    A QTL named Mobe1 (modifier of obesity 1) mapped to 37 cM on mouse Chromosome 2 with peak linkage at D2Mit9 (LOD=9.48). BTBR-derived alleles confer increased 10-week body mass with semidominant inheritance and explains about 14% of the phenotypic variance. Mobe1 was fine-mapped using interval specific congenic strains (ISCS) to an interval flanked by D2Mit56 (41 cM) and D2Mit159 (47.1 cM). This region is syntenic to human Chromosome 2q24-32.

    Mobe2 mapped to 37 cM on mouse Chromosome 13 with peak linkage atD13Mit66 (LOD=8.0). C57BL/6-derived alleles confer increased 10-week body mass with dominant inheritance and explains about 12.5% of the phenotypic variance.

    Mobe3 and Mobe4 mapped to 65 cM on mouse Chromosome 5 (LOD=3.6) and to 34.4 cM on mouse Chromosome 17 (LOD=2.49), respectively. Mobe3 and Mobe4 showed suggestive linkage using interval mapping but the LOD scores improved when multiple interval mapping was employed.
