Notes
  • Experiment
The alcohol withdrawal locus Alcw2 was localized to an interval <1 cM using interval specific congenic strain analysis and recombinant progeny testing techniques. The Alcw2 critical region maps between D4Mit80/Ptprd (37.7 cM/38 cM) and Mpdz/D4Mit115 (38.6 cM/38.7 cM). The congenic strain D2.B6-(D4Mit192-D4Mit327) and the short DNA segment (SDS) strain D2.B6-Mpdz both exhibit significanly less severe alcohol withdrawal compared to DBA/2J mice thus confirming the presence of a gene or genes in the C57BL/6J-derived donor segment, which confers a protective effect on the DBA/2J-derived susceptible background. Mpdz is identified as a viable candidate gene for Alcw2. Sequence analysis revealed 18 polymorphisms between DBA/2J and C57BL/6J, 10 of which result in predicted amino acid changes.
