In order to uncover loci that interact or modify Eae2 on mouse Chromosome 15, a congenic strain was created which introgresses the Eae2 locus from experimental autoimmune encephalomyelitis (EAE) resistant strain RIIIS/J onto the genetic background of EAEsusceptible strain B10.RIII. The congenic segment spans 9.4 cM (D15Mit50) - 19.8 cM (D15Mit182) on chromosome 15. This congenic is referred to as B10.RIII-Eae2RIIIS/J. Unexpectedly, homozygosity for RIIIS/J-derived alleles at Eae2 does not confer EAE resistance in the congenic indicating the effect of Eae2 may be epistatic or dependent on other factors. A genome scan with 111 microsatellite markers was conducted on an (RIIIS/J x B10.RIII-Eae2RIIIS/J)F2 intercross to uncover additional EAE susceptibility loci. A locus on mouse Chromosome 7 named Eae26 was identified with significant linkage to acute disease at D7Mit253 (52.8 cM) with a LOD=3.37. B10.RIII-derived alleles appear to confer disease susceptibility in an additive fashion at Eae26.
