  • Experiment
A QTL approach was used to map 2 previously identified loci involved with taste preference, Qui and Tas1r3 (previously referred to as Sac.) 61 male and 72 female (C57BL/6J x DBA/2J)F2 animals were screened for polymorphic markers on chromosomes 4 and 6 corresponding to the regions of Tas1r3 and Qui, respectively. Parental strain C57BL/6J shows stronger preference for sucrose, saccharin, and quinine solutions as shown by greater intake in a short term 1 bottle test compared to DBA/2J. A marker on mouse Chromosome 4 showed strong linkage to sucrose intake (LOD=8.85) and saccharin intake (LOD=10.61) at D4Mit42 (approximatlely 81 cM), mapping very close to Tas1r3. The C57BL/6J-derived allele appears to increase sucrose and saccharin intake in a dominant fashionat this locus. On mouse Chromosome 6, marker D6Mit338 (approximately 62.3 cM) was significantly linked to quinine intake (LOD=4.55) and maps close to Qui and Prp. C57BL/6J-derived alleles appear to increase quinine intake in an additive fashion at this locus.
