    44 liver epithelial and hepatic carcinoma cell lines derived from (C57BL/6J x C3H/HeJ)F1 and (C3H/HeJ x C57BL/6J)F1 mice were screened for loss of chromosomal regions associated with cell line establishment. Loss of heterozygosity (LOH) at Chromosome 12(43.8% LOH at D12Mit217, D12Mit158, and D12Mit263) and 4 (56.3% LOH at D4Mit13; 53.1% at D4Mit308) were prevalent. Preferential loss of C57BL/6J alleles on Chromosome 12 and 4 (P<0.01) appears to be associated with cell line establishment in both crosses. The locus on Chromosome 12 was resolved to 2 distinct loci, Scrd1 at 6 cM and Scrd2 to the 5 cM range between D12Mit79 (53 cM) and D12Mit20 (58 cM). Scrd1 has synteny to human Chromosome 2p while Scrd2 has synteny with human Chromosome 14q32. LOH in thesehuman chromosomal regions has been reported in various cancers. Other loci also observed to have LOH coincident with cell line establishment map to Chromosome 1 (12.5%), Chromosome 8 (12.5%), Chromosome 11 (15.6%), and Chromosome 14 (21.9%).
