G86R Sod1 is a mutated gene causing ALS in a FVB/N background. G86R Sod1 FVB/N mice were mated to mice of a mixed genetic background involving C57BL/6J and 129X1/SvJ. Pedigree analysis indicated that a number of offspring were protected from the effects of the mG86R mutation. These protected mice were from C57BL/6J and 129X1/SvJ backgrounds, which initiated a genome wide scan for genes that modify the effect of the mutation. A significant modifying effect in male animals was associated with mouse Chromosome 13 between markers D13Mit36 (53 cM) and D13Mit76 (61 cM) with a LOD score of 5.07 observed at marker D13Mit227 (59 cM). This locus is named Sodm1 (Superoxide dismutase 1, soluble, modifier). Linkage was also observed in female animals to a lesser degree (LOD=3.82). Potential candidate genes for Sod1m are Smn1 (54 cM), Birc1a (54 cM, formerly Naip), and Serf1 (53 cM).
