    Authors used an intersubspecific backcross between MSM and DBA/2J mice to map D15Mit151 distal to D15Mit197. Subsequent studiees using BAC and YAC clones revealed that D5Mit110 was mapped between D5Mit197 and D5Mit151 and linked to the gad gene in the backcross panel between gad and C57BL/6J mice. using D5Mit110 as a probe, sveral BACs were cloned from a BAC library. Exon trapping and subsequent computer-based analyses revealed the presence of sequences similar to human and rat Uchl1. A full length mouseUchl1 cDNA clone was isolated. PCR analysis of Uchl1 cDNA revealed alteration in genomic organization in gad mice. A 3.5kb segment including exons 7 and 8 of the Uchl1 gene was deleted in gad mice. Western blot analysis provided further evidence that this alteration in the Uchl1 gene caused the mutation in gad mice.
