BALB/c derived NK cells were found to target Chinese hamster ovary (CHO) cells less efficiently than NK cells derived from the C57BL/6J strain. Analysis of seven recombinant inbred (RI) mouse strains derived from BALB/cBy and C57BL/6By progenitor strainsindicated that roughly 50% of the RI strains displayed either the high or low killing phenotype. This was consistent with the thought that a single chromosomal locus might determine killing of CHO targets. Comparison of the strain distribution pattern forKlra4 with patterns for other genes previously typed in the CXB RI panel revealed complete concordance between Klra4 and Klra1 (author calls it Ly49a), Klrb1c (authors calls it Nkrp1), and Klra8, mapping to distal mouse Chromosome 6. Analysis of the BALB.B6-Cmv1r congenic strain (which possesses the Cmv1r allele and other NK gene complex loci from C57BL/6J backcrossed onto the BALB/c background) confirmed this mapping by demonstrating efficient lysis against CHO cells. The Klra4 gene was later found to be responsible for the phenotype (see J:55124).
