The Ft (fused toes) mutation has been previously mapped to mouse Chromosome 8. In order to localize candidate genes in the region, transgene preintegration site probes were used to isolate regional cDNA clones. Fts was identified as a candidate gene. Direct sequencing of the 3' portion of Fts clones indicated a sequence identical to the 3' reverse complementary sequence of Rbl2. The genes are closely linked, transcribed in opposite directions and polyadenylated transcripts overlap in their 3' UTRs. To confirm the Fts/Rbl2 association authors used PCR primer pairs derived from Fts and Rbl2 sequences to successfully amplify products. Expression studies indicate that Fts is disrupted in the Ft mice but the authors suggest multiple genes are likely to be involved.
