Caption Histological analysis of 6.5-dpc embryos from Sox2tm1.1Vep/Sox2+ (Sox2betageo2/+), Sox2tm1.1Vlcg/Sox2+ (Sox2COIN/+), Sox2tm1.2Vlcg/Sox2+ (Sox2INV/+) intercrosses. Wild-type (A), Sox2tm1.1Vep/Sox2+ (Sox2betageo2/+) (B), Sox2tm1.1Vlcg/Sox2+ (Sox2COIN/+) (D), Sox2tm1.1Vlcg/Sox2tm1.1Vlcg (Sox2COIN/COIN ) (E), Sox2tm1.1Vlcg/Sox2tm1.1Vep (Sox2COIN/betageo2) (F), and Sox2tm1.2Vlcg/Sox2+ (Sox2INV/+) (G) are all phenotypically normal. In contrast, Sox2tm1.2Vlcg/Sox2tm1.2Vlcg (Sox2INV/INV) (I) and Sox2tm1.2Vlcg/Sox2tm1.1Vep (Sox2INV/betageo2) (H) embryos form disorganized extraembryonic tissues, but fail to form the epiblast, effectively phenocopying Sox2tm1.1Vep/Sox2tm1.1Vep (Sox2betageo2/betageo2) embryos (C), and displaying the phenotype previously described for Sox2 homozygous-null embryos. Sections were stained with hematoxylin and eosin (H&E).
