Caption Aberrant embryonic development in Syf2Gt(PST25562)Mfgc/Syf2Gt(PST25562)Mfgc (GT/GT) embryos. The following results were repeated with three independent experiments. (A) Morphology of embryos cultured in vitro. Embryos were collected from uterus at E3.5 and cultured in vitro to E6.5. Embryos were hatched at E6.5 in wild-type (+/+) and heterozygous (GT/+) embryos. Homozygous (GT/GT) mutant blastocysts exhibited a shrink shape without hatching. (B) Embryos were isolated from E7.5 to E9.5 for hematoxylin-eosin staining. Wild-type and heterozygous embryos at E7.5 had normal gastrulation. Homozygous embryos did not undergo normal gastrulation at E7.5. Mutant embryos at E8.5 and E9.5 displayed severely impaired embryonic development of head, trunk, and appendages. ee:embryonic ectoderm. m: mesoderm. ve: visceral endoderm.
