Caption Generation of the Pnpla3tm1Eek allele. (A) The native (wild-type, WT) Pnpla3 allele and targeting (knockout, KO) construct (not drawn to scale). Bacterial artificial chromosome (BAC) recombineering was used to replace 1939 bp of the native Pnpla3 gene from position -12 to +1927 including the ATG start signal and the entire exon 1 (which contains the GXSXG motif with the putative critical catalytic serine residue of the patatin domain) by a neomycin resistance cassette. Embryonic stem (ES) cell electroporation, selection, and screening were then performed using standard techniques.
