(A) Schematic representations of the wild-type Trim3 allele (Top), the targeting construct showing the replacement of most of coding exons 2-5 with a neomycin resistance cassette and the introduction of an additional NcoI site (Middle), and the mutant allele (Trim3tm1Mak) after successful homologous recombination (Bottom). The 5'-flanking probe (fp) located external to the targeting construct was used to identify successfully recombined ES cell clones as well as to confirm mouse genotypes by Southern hybridization. Arrowheads a/c and b/c indicate the primers used for PCR in B.
