The entire open reading frame (ORF) is in one exon. (Middle) Constructed targeting vector, which contains a replacement neomycin-resistance gene, is shown. (Bottom) Predicted genomic structure of the homologous recombined locus is shown. Restriction enzyme sites indicated are Apa I (A), Ban III (B), Nco I (N), and Sal I (S). Southern blot analysis yielded a 4.8-kb Nco I-digested band corresponding to the WT allele and a 3.4-kb Nco I-digested band corresponding to the targeted (B3gnt5-/-) allele, using an external probe (bold line). Designated primers for genotyping PCR are indicated by arrows. Probe indicates the binding region of the DNA probe used for Southern blot analysis.
