Caption Generation and characterization of P1c-deficient mice, Plec/Parp10tm1.1Gwi. A, Schematic representation of the relevant plectin gene locus and the targeting vector. Noncoding exons (1, 0, and 0a) and coding exon (1c, 1f) are represented by gray and black boxes, respectively. Note, the 3 non-coding exons were replaced, along with exon 1c, by a neomycin resistance cassette. Digestion of genomic DNA with the restriction enzyme BamHI releases a 8.6 kb fragment from the wt allele, and a 6.6 kb fragment of the mutant allele. Both fragments are detectable with the 3 external probe shown. Relevant restriction sites are indicated: B, BamHI; H, HindIII; N, NcoI; X, XbaI.
