Caption Generation of the Pphln1Gt(A043F08)Wrst allele. Mapping of the gene trap vector integration site in intron 6 of the murine periphilin-1 gene. The gene trap principle is based on the random integration of a reporter/selector cassette into a genome in order to simultaneously select for, mutate, and identify the trapped locus. Gene trap vectors are promoterless, and the presence of splice donor or acceptor elements leads to the generation of a fusion transcript with the endogenous gene after intron integration. For ES cell clone A043F08, the vector integration site could be mapped to intron 6 of the murine periphilin-1 gene in between exons 6 and 7. Because exon 6 is skipped by alternative splicing, the reporter/selector cassette lies directly downstream of exon 5 in the resulting fusion transcript. Exon 1 is not part of the periphilin-1 coding sequence (CDS), because the start codon lies in exon 2.
