Caption Generation of the Cd209dtm1.1Cfg allele. (a) Schematic representation of the targeting strategy. A targeting vector was constructed to flank a 5.1-kb PCR-amplified Bam HI fragment, (red) including exons 1 to 5 of the Cd209d (SIGNR3) gene, with two loxP sites. Another loxP site was inserted to flank TK and PGK-Neo. A PCR-amplified 2.0-kb Sac II fragment (green) and a 4.0-kb KpnI-Eco RI fragment (blue) were used as 5' and 3' homologous regions, respectively. ES clones with floxed-TK-Neo alleles were selected. Cre-mediated excision was performed to generate floxed and delta (Cd209dtm1.1Cfg) alleles.
