Exon 1 is flanked by two loxP sites, and the neomycin gene cassette is inserted 3' to exon 1 and internal to the distal loxP site. The neomycin gene cassette is bound by two frt sites and includes an additional loxP site. The target region is 0.9 kb and includes exon 1.
