Caption The "RasE Multi-Hit BAC" transgene construct. Overall structure (top) and individual expression modules (bottom). Each expression module is in the inverted (off) position relative to its CAG promoter (P); the expressed sequence is flanked by loxP sites (triangles), oriented head-to-head, so it can be activated by Cre recombinase-mediated "flipping." Deletion of sequences between similarly-oriented loxP sites is prevented by use of incompatible, heterotypic loxP sites for different modules: black represents canonical loxP, blue lox5171, and red lox2272. Other components: FRT, FRT recombinase recognition site; 2xHS4, chicken 2xHS4 insulator sequence; pA, SV40 polyadenylation signal; Gtx, internal ribosomal entry site; eYFP, dsRed, and hCD2t, fluorescent protein reporter genes; S35, G37, and C40, the MAPK, RALGEF and PI3K pathway-activating HrasG12V mutant alleles.
