Caption Construction of the Angptl4 E40K point mutation gene targeting vector to generate the Angptl4tm2Lex allele. (A) A schematic representation of the Angptl4 locus is shown, including relevant restriction sites used for cloning and confirmation Southern hybridization analysis (BamHI, SfiI, BclI, and HindIII). The targeting vector was modified from the previously published Angptl4 conditional targeting vector. The new 5' arm of homology with the required mutation was amplified by PCR. This cDNA and the LoxP-neo-LoxP selection cassette were subcloned into the conditional targeting vector, replacing the old 5' arm of homology and selection cassette. The 5' internal and 3' external probes used for Southern hybridization analysis were designed inside and outside the target vector arms of homology accordingly and were generated by genomic PCR.
