Caption (a) The Skt (Etl4) gene consists of 19 exons and is located in the proximal region of chromosome 2. (b) In the Etl4Gt(pU8)80211Imeg allele, the pU-Hachi (U8) trap vector, comprised of a mutated loxP (lox71), internal ribosomal entry site (IRES), b-geo, poly(A) sites, and loxP, is integrated into the 14th intron of the Etl4 gene. Replacement vector includes an exchange cassette containing a mutant loxKR3 site, IRES, cre with nuclear-localization-signal sequences (Ncre), poly(A) sites, an Frt-flanked puromycin resistance (puro) selection marker, and loxP. When the replacement vector is introduced into the Etl4Gt(pU8)80211Imeg ES cells, specific recombination occur between lox71 and loxKR3, generating Etl4tm1(cre)Kymm (c) In the Etl4-cre ES cells, U8 in the Gt(pU8)80211 gene trap is replaced by the exchange cassette.
