| Caption | A strategy used for Map3k12 (DLK) gene excision. The construct used for homologous recombination utilized a phosphoglycerate kinase promoter-driven neomycin selection cassette (PGK-NEO) flanked by Flip recombinase target (FRT) sites as well as LoxP sites that surrounded exons 2-5 of Map3k12. This construct was targeted using homologous recombination, and then exons 2-5 of Map3k12 were excised via Cre-mediated recombination in ES cells before blastocyst injection to generate the 5294468 allele. | ||||
