(A) Immunofluorescence analysis in metaphase II arrested oocytes (a, f and k) and at different times after fertilization. The embryos were collected for the immunofluorescence at intervals of 2 h. Time was measured starting from hCG injection. (f-j) staining of DNA with Hoechst 33342 dye. (B) Western blot analysis in MII arrested oocytes (n = 100), PN (pronuclear) zygotes (n = 100) or two-cell embryos (n = 75). (C) Western blot analysis in MII arrested oocytes (n = 250) untreated or treated with alkaline phosphatase (AP). (D) Immunofluorescence analysis. (h-n) staining of DNA with Hoechst 33342 dye. (E) Immunofluorescence analysis of mouse early blastocysts with preimmune IgGs (top panels). Bar is 50 um. (F) Western blot analysis in two-cell embryos (n = 50), morulae (n = 25) or blastocysts (n = 25). All data shown are representative of at least three experiments that gave similar results.
