(A) Freshly isolated keratinocytes plated at the same density on a mixture of collagen I and fibronectin. Although control cells are well spread, Itgb1^tm1Ref/Itgb1^tm12.1Ref Tg(KRT5-cre)5132Jlj/0 (YY/AA) mutant cells do not spread and adhere properly. Cells at the indicated time points after seeding are shown. Bar, 25 um. (B) Keratinocytes were cultured for 2 days on a mixture of collagen I and fibronectin and immunostained for Itgb1, talin, vinculin (red), and F-actin (phalloidin; green). Nuclei were stained with DAPI (blue). Itgb1^tm1Ref/Itgb1^tm12.1Ref Tg(KRT5-cre)5132Jlj/0 cells fail to develop focal adhesions and to organize the actin cytoskeleton.
