Histology of soleus muscle from Ryr1^tm1.1Dhm/Ryr1⁺ mice. (A and B) Hematoxylin Eosin stained transverse sections of soleus muscle from 6 month old wild-type (WT, A) and Ryr1^tm1.1Dhm/Ryr1⁺ (IT/+, B) mice. The mutant muscle sample (B) shows increased fiber size variability, rounded fiber outline, and mild endomysial fibrosis. The level of central nucleation is comparable to that in the control sample. Arrowheads show endomysial spacing that is slightly increased in mutant muscle sample. Double-headed arrows in B show high variability in myofiber diameters in mutant muscle. (C and D) NADH-TR stained transverse sections of soleus muscle from 6 week old WT (C) and mutant (D) mice. A similar distribution of lightly stained type 2 (fast) fibers and darkly stained type 1 (slow) fibers is observed in WT (C) and mutant (D) muscles. (D) Increased fiber size variability is an early feature of mutant muscle, affecting both type 1 and type 2 fibers. Type 2 fibers in young mutant mice show a tendency for increased fiber diameters, whereas type 1 fibers (arrowheads) show occasional decrease of fiber diameter (hypotrophy). (Scale bars, 25um).
