Caption Normal wild type (a,c and e) and Upk3atm1Tsun/Upk3atm1Tsun mouse bladders (b,d and f), all from 2-3 month old mice, were examined using hematoxylin and eosin-stained paraffin sections (a and b), or using toluidine blue-stained plastic thin sections (c and d). Appearance of methylene blue-stained wild type (e) and mutant (f) mouse bladders. G: Retention of methylene blue by wild type (n=7) and mutant mouse (n=5) bladder (P=0.019). The dye that was retained by the bladder was solubilized with chloroform and read at 660 nm. Note the rugged luminal surface of the normal, superficial umbrella cells (arrows), and the markedly different mutant urothelium, which was characterized by a smooth, apical cell surface (arrowheads), the absence of an umbrella cell layer, and the leakiness/retention of methylene blue.
