Lrrtm4 expression is detected by whole-mount in situ hybridisation on 8.5 dpc mouse embryos (A) in caudal mesoderm. B is a posterior view, interference contrast image of the embryo in A. Expression in caudal mesoderm (cm) continued at 9.5 dpc (C) with further sites of expression appearing in the neural tube (nt), ventral midbrain (vm), head mesoderm (hm) and limb buds (lb). Specific midbrain expression (D) was also located in the lamina terminalis (lt) that continued at 10 dpc (E). Limb bud (lb) expression was present in the forelimbs (F) and hindlimbs (G). A section through the forelimb (H) shows expression is located in the limb mesenchyme (lm). Transverse section of the tail (I) shows caudal mesoderm (cm) expression is located ventral to the presomitic mesoderm (psm). Neural tube (nt) expression is restricted to ventro-lateral cells dorsal to the floor plate (fp). Expression is also present in head mesoderm (hm) between the forebrain (fb) and hindbrain (hb), either side of Rathke's pouch (K). The approximate plane of sections I, J, and K are shown in lower case (C). At 11 dpc (L) expression appears along the neural tube (nt). In the rostral neural tube expression is confined to lateral ventral cells (M) dorsal to the floor plate (fp). More caudally, expression in lateral cells extends dorsally (N). At the interlimb level, expression is restricted to lateral cells in the centre of the neural tube (O) with expression absent from the neural tube in the tail (P). Expression in the caudal mesoderm occurs ventral to the neural tube (P). The approximate plane of sections M-P are indicated in lower case (L). Further stripes of expression are seen in the caudal half of 10.5 dpc mouse embryos (L). Transverse sections (O and S) show that this expression is localized between the neural tube and dermomyotome, in a subset of the sclerotome (scl). In younger somites this expression is broad and then restricts to the anterior half as the somite develops (Q). In more mature somites expression is localized to a domain in the posterior (post.) half of the somite (R). Limb expression at 11 dpc had decreased however at 10.5 dpc a wedge of expression was still present (T) in the hind limb bud (lb) which was localized to the limb mesenchyme (S).
