Caption (A) Schematic representation of the transversal sections at the foregut region of an E8 mouse embryo (black line in upper panel), and the discernible structures in such sections (lower panel). Transversal sections of an E8 embryo (eight somite pairs) (B-E). The foregut endoderm in the sections in (B) and (D) were visulized by co-staining with the endoderm-specific anti-HNF3b antibody (red), and the sections were subsequently stained with DAPI (blue) (C and E, respectively). The blue staining from the BMP4-specific hybridization in (B) was pseudo-colorized to green in (C). Arrowhead indicate the mesenchyme adjacent to endodermal cells of the liver diverticulum (arrow). (F) Schematic representation of the transversal section of the area at the liver bud of an E9.0 mouse embryo (black line in upper panel), and the relevant structure in such sections (lower panel). In situ hybridization on consecutive transversal sections of an E9.0 liver bud (16 somite pairs, sp). Transversal sections of the liver bud at various E9 stages; 16 sp (I, J), 22 sp, (K, L) and 25 sp (M, N) hybridized with an Lhx2 RNA probe (I, K and M) and subsequently co-stained with an anti-HNF3b antibody (red) (J, L and N). The blue staining from the Lhx2-specific hybridization in I, K and M has been pseudo-colorized to green in J, L and N. Delaminated hepatoblasts beginning to migrate as cords into cells of the mesenchyme can be seen in L (arrows). (O) Schematic representation of the sagittal section through an E9.5 mouse embryo (black line in left panel), and relevant structure in such section (right panel). Combined double in situ hybridization and immunohiostochemical analysis of a sagittal section (P) and a magnification of the squared area (Q). This section was hybridized both to a BMP4 RNA probe (magenta/brown) and an Lhx2 RNA probe (blue), and subsequently stained with an anti-HNF3b antibody (pink/light red). fg; foregut endoderm; he; heart; lb; liver bud; ld; liver diverticulum; nt, neural tube; st; septum transversum mesenchyme. Scale bar, (B-E and Q) 100 microns; (G-N) 50 microns; (P) 200 microns.
