Caption (A-D) Wild-type embryos. (E-L) Mutant embryos. (B) Higher magnification of the tail region after clearing of the embryo in A. (C) A 12.5-day wild-type embryo and (D) its tail region after clearing. broad arrow, notochord anterior to the base of the tail. The tailbud is smaller than at 11.5 days. (E) At 10.5 days no external difference can be detected between wild-type and heterozygous mutant embryos. (I) After clearing, however the mutant is phenotypically distinguishable from wild-type litter mates. The notochord of the posterior trunk region is disrupted posterior to the hindlimb buds. (F) At 11.5 days, heterozygous TWis embryos show a constriction (upper arrow) between tail and trunk. The neural tube of this particular embryo is not closed at the sacral level (lower arrow). (J) The notochord in the posterior trunk and anterior tail region is fragmented and often bifurcated (not shown). (G) At 12.5 days TWis heterozygotes show a thinning of the tail posterior to the constriction (arrow). (H) At 13.5 days the axial structures of the tail have disintegrated (arrow) to an unstructured appendix which is often bifurcated (L). (M-P) Alizarin-stained partial skeletons of 18.5-day embryos. (M) Wild-type 18.5-day embryo; (N-P) TWis/+ heterozygotes showing a variable severity of skeletal malformations. Arrows highlight malformations of vertebrae and ribs, fusions of dorsal arches, and absence of vertebral bodies or vertebrae. Even thoracic segments are occasionally affected (P). nc, notochord; tb, tail blastema. Scale bars: in A,F, and D, 550 um; B,I,J,K, and L, 400 um; C,G, and H, 1.4 mm; N,O, and P, 1.7 mm.
