Caption (A-F) Transverse sections from posterior to anterior through an 8.5-day whole mount wild-type embryo (corresponding to planes 1 to 6 on Fig. 3D). In the most posterior section (A) a mass of mesodermal cells underlies the embryonic primitive ectoderm layer of the primitive streak. The notochordal plate is first visible in (C). (C-E) In relation to particular cross sections the number of cells of the notochord as well as the size of the neural plate decreases during differentiation, while the paraxial mesoderm is condensing to somites. (F) The notochord is only two to three cells wide. (G-I) An anterior-posterior series of sections through an 8.5-day mutant embryo (corresponding to planes 1-3 on Fig. 3H). In (H) a remaining notochordal plate cell is found approximately at the posterior end of the initially formed notochordal plate. Somites are not formed (broad arrow), but the lateral plate mesoderm is present. In the posterior primitive streak region (I) cell proliferation has continued in the absence of mesoderm formation; a bulk of densely packed cells has formed (broad arrow). ee, primitive embryonic ectoderm; lpm, lateral plate mesoderm; m, mesoderm; ncp, notochordal plate; np, neural plate; nc, notochord; nf, neural fold; nt, neural tube; so, somite. Scale bars: A-H, 75 um; I, 150 um.
