behavior/neurological
N |
• mice exhibit a normal response to agonist U50,488-induced analgesia relative to control mice, as shown by similarly increased tail withdrawal latencies in the warm water tail-flick assay
• baseline tail withdrawal latencies are not significantly altered in the tail-flick assay
• mice exhibit significantly greater locomotor sensitization to cocaine (15 mg/kg) than control mice at day 8 of testing
• however, no differences are detected in a cocaine conditioned place preference paradigm following conditioning with 5 mg/kg or 1.25 mg/kg cocaine
• mice spend significantly more time than control mice in the center of the open field during the final 45 min of the test session, indicating an anxiolytic-like effect
• mice show significantly shorter latencies to first enter the light side of the light/dark box, with no differences in total time spent in the light compartment relative to control mice
• however, no differences are detected in open arm time or open arm entries in the elevated plus maze test

nervous system
• cocaine-induced synaptic strengthening in the ventral tegmental area does not occur in mice after cre activity is induced
• NMDA-mediated excitatory postsynaptic currents are completely abolished in the ventral tegmental area of mice after cre activity is induced
behavior/neurological
• mice that have had cre activity induced and were conditioned to associate a certain cage environment with cocaine do not reinstate their cocaine-seeking behavior when administered the drug after a 16 day extinction phase

behavior/neurological
• mice do not lose the preference for an environment where cocaine was previously administered (i.e. conditioned place preference) even after 16 days have passed since the last cocaine dosing
nervous system
• there is no increase in the ratio of AMPA-/NMDA- mediated receptor currents for the ventral tegmental area isolated from mice treated with cocaine as occurs in wild-type controls
• cocaine-induced synaptic strengthening in the ventral tegmental area does not occur in these mice

nervous system
• basal glutamate-mediated receptor currents are enhanced in these mice
• cocaine-induced synaptic strengthening in the ventral tegmental area does not occur in these mice
