Galectin-8 markedly enhances VEGF-C-induced lymphangiogenesis in vivo in Tg(Prox1-EGFP)KY221Gsat/0 mice

immune system
• in corneal micropocket assays, corneas implanted with both VEGF-C and galectin-8 pellets show enhanced VEGF-C induced lymphangiogenesis relative to corneas implanted with either VEGF-C or galectin-8 pellets alone, as calculated by the lymphatic vessel area on day 7 post-surgery
• after suture placement or silver nitrate cauterization to induce corneal inflammation, treatment with TDG (a pan inhibitor of galectins) or Gal-8N (a dominant negative inhibitor of galectin-8) significantly suppresses both suture- and cautery-induced inflammatory lymphangiogenesis, as quantified by the lymphatic vessel area at the end of the treatment period
• in corneal micropocket assays, corneas implanted with either VEGF-C or galectin-8 pellets exhibit >35% of proliferative lymphatic endothelial cells (Prox1+ Ki67+ LECs) in lymphangiogenic areas, whereas corneas implanted with control pellets show <4% of Prox1+ Ki67+ LECs on day 7 post-surgery
