• at 2 weeks after transverse aortic constriction (TAC) surgery, mice show preserved cardiac function with a significantly higher ejection fraction (EF) and fractional shortening (FS) and a significantly lower left ventricle weight to body weight (LV/BW) ratio than TAC-operated wild-type controls
• at 4 weeks after TAC, both EF and FS are significantly higher while mRNA levels of Nppa and Nppb (fetal stress markers0 are 1.9-fold and 3-fold lower, respectively, than in TAC-operated wild-type controls, indicating protection against pathological remodeling in response to pressure overload
• 4 weeks after TAC, mice show a ~2-fold decrease in GRK5 protein levels but no change in Grk5 mRNA levels in the left ventricles (LVs); GRK5 protein levels are blocked in nuclear but not in cytoplasmic fractions from LV lysates
• pathological remodeling is decreased as early as 2 days after surgery: TAC-induced Nppa and Nppb mRNA levels and induction of MEF2A (myocyte enhancer factor 2A) target genes Xirp1 and Nr4a1 are significantly decreased; LV/BW ratio and GRK5 protein levels in LVs are significantly lower whereas protein levels of neddylated CUL1 are higher than in TAC-operated wild-type controls
• however, baseline cardiac function is normal as assessed by EF and LV/BW ratio, with no significant change in Cand1, Nppa or Nppb transcript levels relative to wild-type controls
