Altered size of secretory lysosomes in Ebag9tm1Rehm/Ebag9tm1Rehm cytotoxic T lymphocytes

immune system
• lytic granules in CD8 + T cells have a 30% decrease in volume
• CD8 + T cells have 3-fold more cytolytic activity than controls in an allogenic cytotoxicity assay
• this enhanced cytotoxicity results from increased release of granzyme-containing lytic granules
• granzyme B is sorted to secretory lysosomes more efficiently than controls
• CTLs eliminate twice as many targets as controls when TAG-loaded splenocytes are transferred into mutant mice
• significantly lower Listeria monocytogenes titers are found after infection also indicating enhanced CTL activity
• there is an enhanced secondary CD8 + T cell response to antigen loaded splenocytes after secondary challenge with antigen
• bacterial titers of Listeria monocytogenes are more than a log unit lower than controls 6 days after infection
