• T-helper 2 cell differentiation and cytokine production is impaired following stimulation with anti-CD3 antibodies with or without anti-CD28 antibodies, phorbol-12- myristate-13-acetate with ionomycin, or strong TCR-mediated suboptimal Th2 polarization regardless of the presence of absence of exogenous IL4
• 2 weeks following the second immunization with ovalbumin, mice produce 2-fold less ovalbumin-specific IgG1 compared to similarly treated wild-type mice
• 24 hours after stimulation with anti-CD3 antibodies with or without anti-CD28 antibodies, IFN-gamma production is moderately increased compared to similarly treated wild-type cells
• when stimulated with strong TCR-mediated suboptimal Th2 polarization regardless of the presence of absence of exogenous IL4, IFN-gamma production is moderately increased compared to similarly treated wild-type cells
• however, IFN-gamma production after 72 hours of stimulation with anti-CD3 and anti-CD28 antibodies or optimal TCR signaling is normal
• 24 hours after stimulation with anti-CD3 antibodies with or without anti-CD28 antibodies or phorbol-12- myristate-13-acetate with ionomycin, CD4+ T cells exhibit decreased IL13 production compared to similarly treated wild-type cells
• after stimulation with strong TCR-mediated suboptimal Th2 polarization with exogenous IL4 expression, IL13 production is less than in similarly treated wild-type cells
• however, IL13 production after 72 hours of stimulation with anti-CD3 and anti-CD28 antibodies or optimal Th2 conditions is normal
