growth/size/body
• 4-week old mice have an average spleen weight 2-fold higher than that of the littermate controls
• however, by 8 weeks of age spleen sizes are similar to age-matched controls

hematopoietic system
• 4-week old mice have an average spleen weight 2-fold higher than that of the littermate controls
• however, by 8 weeks of age spleen sizes are similar to age-matched controls
• a doubling of CMP bone marrow cells (Lin-CD34+FcgammaRmed) are found within the myeloid progenitor cell population
• however, transfer of mutant bone marrow to wild-type mice results in a failure of CFU-S colonies to form in the spleen due to proliferation defects
• mice become moribund and die 5 days after hemolytic anemia is induced while control mice all live
• mice fail to increase spleen size after induction of hemolytic anemia
• the red blood cell count and hematocrit are severely reduced in comparison to controls after induction of hemolytic anemia
• decreased B220+ cells are found in the bone marrow with about a 25% reduction compared to wild-type

• excess myeloid progenitor cells are found by colony formation assays at this age
• by 8 weeks of age, extramedullary hematopoiesis appears to have stopped
• a mild anemia is observed in 4-week old mice
• fewer Ter119+ erythroid progenitor cells are found in the bone marrow
• only a few BFU-E form in colony assays
• when hemolytic anemia is induced, mice fail to increase the number of BFU-Es in the spleen as controls do
• there is more than a 50% decrease in MEP bone marrow cells (Lin-CD34-FcgammaR-) that are found within the myeloid progenitor cell population
• reduced numbers of CD41+ cells are found in the bone marrow
• there is an increased number of Howell-Jolly bodies found in circulating blood cells
• mean reticulocyte percentage in the blood of 4-week old mice is 16% versus 8% in wild-type mice
