immune system
• alpha beta lineage T cell undergo increased apoptosis compared to in wild-type mice
• thymic cellularity is reduced to 1% of normal
• differentiation of alpha beta-lineage precursors into more mature double and single positive T cells is severely impaired and is arrested at beta selection
• differentiation of alpha beta-lineage precursors into more mature double and single positive T cells is arrested at beta selection
• arrested alpha beta T cell development is dependent on Trp53 expression
• however, alpha beta lineage T cell development is cell autonomous and restored by ectopic expression of Rpl22
• alpha beta lineage T cells fail to differentiate to DN4 cells
• while gamma delta T cell lineages are increased in proportion and absolute number, alpha beta T cell lineage proportion and numbers are decreased in lymphoid organs
• fewer CD44- CD25- DN4 T cells are present
• decreased T cell numbers are associated with increased apoptosis
• mice exhibit a reduction in both DN3 and DN4 T cells
