phenotype observed in females
phenotype observed in males
N normal phenotype
mortality/aging
• diabetes-like symptoms (dehydration, weight loss) lead to death of most animals by 6 months of age

growth/size/body

immune system
• double mutants develop insulitis at 2 months of age
• mice have a greater number of cytokine-positive cells (Il4 or Ifng) compared to mice coexpressing CD80 and the MHC class II transgene
• mice develop clinical signs of diabetes

endocrine/exocrine glands
• mice show autoimmune destruction of pancreatic beta cells
• double mutants develop insulitis at 2 months of age

homeostasis/metabolism
• mice show elevated blood glucose levels (363 mg/dl at 8 weeks rising to 402 mg/dl at 6 months)

N normal phenotype
immune system
• transgenic mice do not develop autoimmune diabetes

homeostasis/metabolism
• islets from transgenic diabetic animals stain weakly for insulin compared to controls
• blood glucose levels are ~300 mg.dl at 8 weeks, increasing to 412 mg/dl by 6 months (J:93555)
• transgenic animals show elevated blood glucose throughout life (J:127706)
• diabetes, developed by all transgenic animals, is characterized by onset of hyperglycemia at 3-5 weeks of age
• blood glucose level at 6-10 weeks is 160-1055 mg/dl increasing to 543-1255 mg/dl at 10 weeks compared to 124-223 mg/dl in controls
• levels are >500 mg/dl in most animals by 2 months of age
• serum is grossly lipemic in transgenic mice; some diabetic animals have levels above 120 mg/dl
• urine tests positive for presence of ketones; ketonuria correlates with severity of diabetes

endocrine/exocrine glands
• islets from transgenic diabetic animals stain weakly for insulin compared to controls

renal/urinary system
• urine tests positive for presence of ketones; ketonuria correlates with severity of diabetes
• accompanies rise in blood glucose and polydispsia

behavior/neurological
• at 8 weeks, water consumption is observed to be ~50 ml/day compared to ~4.2 ml/day in controls
