• morphine treated homozygotes are able to tolerate higher morphine doses compared to wild-type mice (LD50 of 800 mg/kg compared to 700 mg/kg for wild-type)

• in previously unexposed female homozygotes reduced analgesia at low morphine doses is seen, a similar but not significant effect was seen in males

• acute morphine administration induces the same degree of antinociception; however homozygotes develop tolerance to morphine more rapidly compared to wild-type mice in the hot and cold plate tests
• spinal mechanisms of morphine tolerance are normal

growth/size/body
• body weight is normal at birth but homozygotes gain less weight and males and females are 22% and 27% smaller than their wild-type littermates at 2 - 3 weeks of age
• After 3 weeks of age homozygotes gain weight more rapidly and the body weight difference is gone by 2 - 3 months of age
