• homozygotes display no defects in humoral immunity, delayed type hypersensitivity (DTH) or CD8+ cytotoxic T lymphocyte (CTL) responses relative to wild-type littermates
• homozygotes display an impaired contact hypersensitivity response (a dendritic cell-dependent T cell-mediated response), due to defects in T cell priming and cytokine production
• upon in vitro restimulation with dinitro-benzene-sulfonic acid (DNBS, a water soluble analog of DNFB), draining lymph node cells isolated from DNFB-sensitized homozygotes show a 50% decrease in cell proliferation relative to wild-type lymph node cells
• upon in vitro restimulation with oxazolone-conjugated wild-type splenocytes, Thy1.2+ T cells, CD4+ T cells, and CD8+ T cells isolated from the draining lymph nodes of oxazolone-sensitized homozygotes proliferate significantly less than similarly-treated wild-type cells
• in vitro, mutant dendritic cells are defective in costimulating T cell cytokine production
• however, mutant dendritic cells induce normal levels of CD4+ T cell proliferation relative to wild-type cells

• following sensitization with either low (0.05%) or high (1.0%) doses of oxazolone, homozygotes show a >40% reduction in ear swelling, relative to wild-type controls, at 48 hrs post-challenge with 1% oxazolone
• following sensitization with 0.1% DNFB or 0.5% FITC, homozygotes exhibit a >60% reduction in ear swelling, relative to wild-type controls, at 24 hrs post-challenge with 0.25% DNFB or 0.5% FITC, respectively
• wild-type mice adoptively transferred with oxazolone (0.05%)-sensitized homozygous mutant lymph node cells show a >80% reduction in ear swelling at 48 hrs after oxazolon (1%) challenge relative to wild-type recipient mice adoptively transferred with sensitized wild-type cells, indicating a defect in the sensitization phase and not during the homing of T cells to the challenge site
• upon in vitro restimulation with dinitro-benzene-sulfonic acid (DNBS, a water soluble analog of DNFB), draining lymph node cells isolated from DNFB-sensitized homozygotes show a 90% reduction in IFN-gamma production relative to wild-type lymph node cells
• when used as stimulators in a MLR with wild-type BALB/c (H-2d) CD4+ T cell responders, mutant 129/Sv (H-2b) dendritic cells stimulate less than 5% of wild-type IFN-gamma production
• when used as stimulators in a MLR with wild-type BALB/c (H-2d) CD4+ T cell responders, mutant 129/Sv (H-2b) dendritic cells stimulate less than 65% of wild-type IL-2 production
• when used as stimulators in a MLR with wild-type BALB/c (H-2d) CD4+ T cell responders, mutant 129/Sv (H-2b) dendritic cells stimulate less than 30% of wild-type IL-4 production
