  Arginine codon 1119 (CGG) in exon 27 was changed to alanine (p.R1119A) and a loxP site flanked neomycin resistance gene cassette was inserted into intron 26. The neo cassette was removed through subsequent Cre-mediated recombination. The mutation, in the nuclear localization signal (NLS) of the encoded protein, blocks its transport to the nucleus in Schwann cells, leading to peripheral hypermyelination. (J:232487)
