Mutation details:
Using CRISPR/Cas9 endonuclease-mediated genome editing. A single guide RNA (5'-GCAAAGGACGGGCAAGCGGA) was used to introduce a conditional by inversion (COIN) cassette into intron 1 of the luteinizing hormone/choriogonadotropin receptor (Lhcgr) gene. The entire construct has an initial inward facing lox2722 site, an inward facing lox5171 site, the inverted cassette containing a rabbit beta-globin splice acceptor (SA), a P2A self-cleaving peptide, an enhanced green fluorescent protein gene (EGFP), and a bovine growth hormone (bGH) polyadenylation signal. The inversion cassette is then followed by lox2272 and lox5171 sites, both in reverse orientation.
