Mutation detailsLysine codon 182 (AAA) was changed to arginine (AGG) (p.K182R in NP_671493.1 or ENSMUSP00000077208) using an sgRNA (equivalent to AGGTTTGGACATGGTTTTCCNGG) and an ssODN template with CRISPR/Cas9 technology. The mutation reduces the transcriptional coactivation activity (with glucocorticoid receptor (GR)) of the encoded protein, but not its corepression function, which leads to increased dexamethasone-induced insulin resistance.
