Mutation detailsThe endogenous stop codon of Ednrb was replaced with T2A-iCre/ERT2, followed by a 3xSTOP sequence (tgactgactgat), endogenous polyA signal, 3' regulatory elements, and FRT-flanked Neomycin resistance cassette, directed by a guide RNA (gRNA) targeting the genomic sequence. Double-stranded donor DNA containing the T2A-iCre/ERT2 construct (an optimized variant of Cre recombinase [iCre; improved with mammalian codon usage] fused to Cre-ER(T2) [a G400V/M543A/L544A triple mutation of the human estrogen receptor ligand binding domain]) and flanking homologous arms was co-injected with gRNA and Cas9 protein into (C57BL/6 x 129S4/SvJae)F1 derived v6.5 mouse embryonic stem (ES) cells. Flp recombinase removed the frt-flanked neo cassette. (J:101977)
