• Mutation details: The targeting construct inserted upstream of exon 2 an FRT site, a splice acceptor, and eGFP reporter gene, followed by a polyadenylation signal, a loxP site, and a neomycin resistance cassette with a 3' FRT site. It also inserted loxP sites surrounding exons 2 and 3. Cre-mediated recombination removed the selection cassette and floxed exons. Targeted RT-PCR confirmed this allele to be null, as it only expresses exon 1 and the GFP reporter. Western blot analysis confirmed the absence of protein expression in P14 retinal extracts from homozygous mutant mice. (J:369940)
