• Mutation details: The targeting construct inserted upstream of exon 2 an FRT site, a splice acceptor, and eGFP reporter gene, followed by a polyadenylation signal, a loxP site, and a neomycin resistance cassette with a 3' FRT site. It also inserted loxP sites surrounding exons 2 and 3. Targeted RT-PCR suggested that this allele transcribes all exons, as indicated by the existence of the last exon E5 transcripts. Western blot analysis confirmed the absence of protein expression in P14 retinal extracts from homozygous mutant mice, probably due to truncation of mRNA translation from the stop codon in GFP or neo.
