Mutation detailsUsing CRISPR/Cas9-techology, an initial loxP site was inserted into intron 24 of the endogenous TATA-box binding protein associated factor 1 (Taf1) locus. After which a marker cassette, and a second loxP site, are followed by an 81-kb fragment of a patient-derived human TAF1 genomic sequence from intron 24 through the 3' UTR, but lacking the XDP-associated SVA_F retrotransposon, were introduced via homologous recombination. The marker cassette includes a human eukaryotic translation elongation factor 1 alpha 1 (EEF1A1) promoter, mNeonGreen sequence, a human mini hypoxanthine phosphoribosyltransferase 1 (HPRT1) gene, T2A self-cleaving peptide sequence, a neomycin resistance gene, and a Simian virus 40 PolyA (SV40pA) sequence. There is also a STOP codon in the marker cassette (in NeoR), as well as the native STOP codon in human TAF1 exon 38. TAF1 human coding exons 25-38 have been engineered as needed to code for a fully mouse TAF1 protein. (J:94077)
