Mutation details: TALEN-mediated genome editing was used to generate a deletion of exon 6, which results in the ablation of the DDI2 protease domain (delta254-296) of the encoded protein. Immunoblot analysis showed that the DDI2ex6 protein is scarcely detectable in tissue lysates from E10.5 homozygous embryos. Biophysical characterization of the DDI2ex6 protein using nuclear magnetic resonance (NMR) showed protein signals of acquired secondary structures; however, the melting temperature of DDI2ex6 could not be measured by differential scanning fluorimetry (DSF), indicating that, in vivo, the DDI2ex6 protein likely undergoes rapid degradation.
