First, a loxP flanked PGK-Neo cassette was inserted upstream of the ATG start in exon 1 and then, a second loxP flanked PGK-Hygro cassette was inserted downstream of the TGA stop exon downstream of exon 26. Cre-mediated recombination deleted exons 1-26 and the selection cassettes. qRT-PCR confirmed the absence of mRNA transcripts in the liver. (J:266699)
