Mutation detailsThe allele was generated by inserting a DIO-SPOTlight construct into the GT(ROSA)26Sor locus (Ai9 Addgene, 22799), replacing the Lox-STOP-Lox-tdTomato cassette. The final targeting construct consists of a CAG (CMV enhancer/chicken beta-actin) promoter, inward facing lox2272 and loxP sites upstream of the activating transcription factor 4 (Atf4) open reading frame 1 (uORF1), an inverted SPOTlight cassette (enhanced green fluorescent (EGFP) cDNA sequence inserted after the upstream open reading frame 2 (uORF2) methionine start codon of the Atf4 gene and a red fluorescent protein (tdTomato) cDNA replacing the Atf4 start codon), followed by a second lox2272 site and second loxP site in opposite orientations to the first, a woodchuck hepatitis virus post-transcriptional regulatory element (WPRE), a BGH polyA signal, and an attB/attP-flanked PGK-FRT-Neo-polyA cassette (J:101977)
